适用于人胚胎干细胞(hESC)和诱导多能干细胞(hiPSC)的无动物源成分悬浮培养基
TeSR™-AOF 3D是一种无动物源成分培养基,专为人胚胎干细胞(hESC)和诱导多能干细胞(hiPSC)在3D悬浮培养中以聚集体形式扩增及规模化培养而开发。基于TeSR™-AOF配方,TeSR™-AOF 3D培养基性能稳定,其优化的分批补给方案适用于悬浮培养中规模化生产人胚胎干细胞(hESC)和诱导多能干细胞(hiPSC)。创新的分批补给流程节省时间和培养基;每日补给可补充营养,无需在非传代日更换培养基。
TeSR™-AOF 3D系统兼容以下基质预培养的细胞:mTeSR™1(产品号 #85850)、mTeSR™ Plus(产品号 #100-0276)、TeSR™-AOF(产品号 #100-0401)或TeSR™-E8™(产品号 #05990),培养基质包括Corning® Matrigel® hESC-Qualified Matrix(Corning 产品号 #354277)、Vitronectin XF™(产品号 #07180)或CellAdhere™ Laminin-521(产品号 #77003)。与多种悬浮培养容器兼容。
该培养基及其组成成分的生产过程中,至少在二级生产原材料水平不含任何动物或人源成分。TeSR™-AOF 3D 在二级生产原材料上无动物成分。每批用于配制TeSR™-AOF完全培养基的TeSR™-AOF 3D接种补充剂和TeSR™-AOF 3D补给补充剂经过性能测试,并在培养实验中使用人多能干细胞进行验证。

Multiple hPSC lines maintained in TeSR™-AOF 3D depict morphology characteristic of hPSC aggregates in suspension culture, namely a mostly spherical shape, clear edges (not smooth or shiny), even color, a dimpled or pockmarked appearance, and are less than 400 µm in size.
